What does the “No Man / Say What?” meme mean?

Where does the “No Man / Say What?” meme come from?
The source video was uploaded to YouTube on November 28, 2009, by user MexicanSunflower. It relies on a scene from the animated series Clone High, aired on MTV and Teletoon. In this clip, the character Gandhi attempts to improvise acting lessons. The editor isolated the line 'Whaaaaaat' and applied pitch-correction to align the notes with music. The terms 'No Man' and 'Say What?' come directly from the dialogue of this specific scene. This montage inspired many response videos using the same vocal remix technique.
